{"id":4517,"date":"2021-05-25T07:40:36","date_gmt":"2021-05-25T07:40:36","guid":{"rendered":"https:\/\/didsabz-co.com\/?p=4517"},"modified":"2021-05-25T07:42:52","modified_gmt":"2021-05-25T07:42:52","slug":"tissue-staining-2","status":"publish","type":"post","link":"https:\/\/didsabz-co.com\/en\/tissue-staining-2\/","title":{"rendered":"Tissue staining"},"content":{"rendered":"<h4 style=\"text-align: left;\"><strong>Tissue staining stage :<\/strong><\/h4>\n<p style=\"text-align: left;\">this step includes 2 types of general and specific methods . the general methods is : Hematoxylin-eosin method , with this method the nucleus turns to purple and cytoplasm turns to pink.<\/p>\n<p style=\"text-align: left;\">The specific method is that each cell component takes a specific color, which includes staining of frozen sections , connective tissue , neural system , staining for some cytoplasmic materials , staining for enzymes , staining for micro organisms and cytological staining.<\/p>\n<p style=\"text-align: left;\">Solutions used in routine staining include : Ethanol, xylene, Hematoxylin , Eosin , Hydrochloric Acid, lithium Carbonate, Distilled water, Acetic Acid.<\/p>\n<h5 style=\"text-align: left;\">Note:<\/h5>\n<p style=\"text-align: left;\">0.5 cc of Acetic Acid should be added to every 100 cc of Hematoxyline before use.<\/p>\n<h5 style=\"text-align: left;\">Note:<\/h5>\n<p style=\"text-align: left;\">Place the sections in the oven at temperature of 120\u25e6 C for 15-20 minutes before staining , letting them stick completely to the slides.<\/p>\n<h5 style=\"text-align: left;\">Procedure:<\/h5>\n<ol>\n<li style=\"text-align: left;\">pure xylene (1-2 minutes)<\/li>\n<li style=\"text-align: left;\">pure xylene (1-2 minutes)<\/li>\n<li style=\"text-align: left;\">pure xylene (1-2 minutes)<\/li>\n<li style=\"text-align: left;\">\u00a0ethanol 96% (2-3 minutes)<\/li>\n<li style=\"text-align: left;\">\u00a0ethanol 80% (2-3 minutes)<\/li>\n<li style=\"text-align: left;\">\u00a0ethanol 70% (5 minutes)<\/li>\n<\/ol>\n<p style=\"text-align: left;\">Rinsed with distilled water( temperature about 36 degree)<\/p>\n<p style=\"text-align: left;\">7.\u00a0 Hematoxylins : 5 to 10 minutes, red<\/p>\n<p style=\"text-align: left;\">Rinsed with distilled water<\/p>\n<p style=\"text-align: left;\">8. 1% alcohol acid (prepare 70% alcohol and pour 1 cc of concentrated hydrochloric acid for every 100 cc of alcohol)<\/p>\n<p style=\"text-align: left;\">9. 1% saturated lithium carbonate: several dips (per 100cc of distilled water, one gram of lithium carbonate powder, blue)<\/p>\n<p style=\"text-align: left;\">10.
